dc.description.abstract | Edible Bird Nest (EBN) is a well-known, highly priced and medicinal product in the food industry. The EBN is known to bring many health benefits. Unfortunately, the high pricing of the EBN is due to the tediousness in cleaning the EBN and the usage of man power just to clean it. The conventional cleaning of EBN involves the usage of tweezers and hydrogen peroxide to remove the impurities. However, bromelain enzyme from pineapple waste seems to be a good alternative to clean the EBN. It is a more environmental friendly and sustainable method compared to the conventional method. In this project, bromelain enzymatic assay was carried out to study the activity of enzyme. Moreover, optimization of enzyme activity was carried out to obtain the highest yield of enzyme activity. Three parameters (temperature of centrifugation, t-butanol to crude enzyme ratio and percentage of ammonium sulphate concentration) were studied using the Design of Experiment (DOE). Three phase partitioning method was used. Proteins from bromelain enzyme was precipitated at the interphase of aqueous phase and organic phase. The DOE readings showed that the increasing butanol ratio to crude enzyme ratio has its significant effect on bromelain activity. The solution obtained from the DOE showed that the bromelain activity is best optimized at parameters, butanol to cude enzyme ratio of 1:1, temperature of centrifugation at 20.74 ˚C and percentage of ammonium sulphate of 61.89%. Besides that, the optimization of bromelain enzyme activity showed a higher yield when the percentage of ammonium sulphate concentration was higher. Enzyme stability test was also carried out based on three different pH values of pH 6, pH 7 and pH 8 and three different temperatures at -25 ˚C, 4 ˚C and 28 ˚C. The data obtained showed that the bromelain enzyme was most stable at temperature 4 ˚C and pH 7. This was done to determine the most suitable condition of enzymes. | en_US |
