Molecular identification of Harumanis mango by DNA sequencing and comparison of the Chloroplast DNA sequence
Abstract
Mangifera indica L. var. Harumanis is a mango cultivar widely known for its sweet taste, aromas and the vibrant of mango flesh colour. The studies on morphological characterization of Harumanis have been abundant throughout the recent years. However, the genetic identification based on genotype of Harumanis mango and the sequences are still lack of studies. The aim of this research is to study on the identification of DNA target sequence of chloroplast DNA using DNA sequencing and in silico analysis. The sample from five selected locations was extracted using CTAB DNA extraction. The extracted genomic DNA and the amplified products of polymerase chain reaction (PCR) were analyzed through gel electrophoresis and gel documentation system. It was found that the best annealing temperature for primer cp104 is 62.4 oC and primer trnL-F is 60.4 oC. The similarity of genetic identification and BLAST results showed that all the 30 samples extracted from this study is 99% identical to the Mangifera indica existed among the Harumanis accessions.
