Total phenolic content is an active secondary plant metabolites which can act as antioxidants and are widely distributed in many foods of plant origin. This research is to optimize the extraction of total phenolic content. The best drying method for recovery of total phenolic content were evaluated. The optimum extraction conditions for total phenolics content of mango byproduct were analysed using response surface methodology. The effects of extraction time (15-60 min), solvent (ethanol) concentration (0-100%) and sample-solvent ratio (1:10-1:50 g/mL) were investigated. Quadratic model were highly significant (p < 0.0001) for the response studied with high coefficients of determination (R2) of 0.9682. The result of study suggests that 52 min of extraction time, 54.16% of ethanol concentration and 1g:47.65mL of sample-solvent ratio are the optimal condition to achieve the highest yield of TPC (69.71 mg GAE/g d.w). The experimental values agreed well with those predicted, indicating the suitability of the model and the success of response surface methodology in optimizing the extraction conditions.en_US
