    Extraction, separation and optimization of Gadis Lompat (Chloranthus Erectus) herb

    Abstract
    The Soxhlet extraction method was used to optimize the extraction yield of Chloranthus erectus leaves. Soxhlet extractions were performed at five varies extraction time (60 minutes, 165 minutes, 270 minutes, 375 minutes, and 480 minutes) with two types of solvents (methanol and dichloromethane). The user defined in response surface methodology with two factors (time and types of solvent) was implemented. The analysis of variances (ANOVA) showed the extraction time was no significant while the type of solvent factor was a significant factor. The study showed that methanol extract was almost four times than dichloromethane extract. The highest yield of methanol was 15.317 % compared to the yield of dichloromethane which was just 3.430 %. DPPH free radical-scavenging found that dichloromethane solvent showed higher percentage (60.49 %) compared to the methanol extracted yield (59.51 %). Thin layer chromatography showed the extracted yield of Chloranthus erectus had good separation in the mobile phase of the methanol- acetic acid mixture (containing 99.5% (v/v) methanol and 0.5% acetic acid). The mixture was mixed with water at a ratio of 9:1 (v/v). This mobile phase was able to separate four visualized components for methanol extract and three visualized components for the dichloromethane extract. By using the same mobile phase, the HPLC showed that there were total 34 peaks which indicated 34 unknown compounds contained in the methanol extract of Chloranthus erectus .While HPLC showed that there were total 25 peaks which indicated there are 25 unknown compounds contain in the dichloromethane extract of Chloranthus erectus.
