    Optimization of antioxidant application in tissue culture of Harum Manis (Mangifera indica L.) for browning

    Abstract
    Harum Manis mango, a variety of the mango fruit Mangifera Indica L., is said to be one of the best varities in the world. In mango, successful micropropagation through plant tissue culture has been achieved through somatic embryogenesis from nucellar tissues of immature ovules. The in vitro propagation of plantain Harum Manis mango countered with many problems and one of them is browning or blackening of tissues prior to culture due to oxidation of phenolic compound by polyphenolic oxidase enzyme present in the tissue when excised. An investigation purposely to catter this problem by using pamelo peel extract was conducted. In this study, volume of pamelo peel extracts, time of subcultures and condition of dehandling were factors identified caneffectively reduced the browning occurrence in mango tissue culture. The results for this study, 200 mL of pamelo peel extract had delayed the browning occurrence which is at 180 minutes. When the volume was high the time of browning to happen was longer. Condition dehandling method excised explants under water give the best results shows the longest time to explants turn to browning it take more 30 minutes than cutting in the air. Time of browning to occur will also take longer if there are subcultures. If not done subculture time of browning will taken shorter. The result shows the time of subculture before or after it will not give effect to the time of browning of the explants.
