Effects of substrate composition on mycelium growth and yield of Schizophyllum Commune, S.commune was studied. Specifically, this type of mushroom is a kind of mushroom that belongs to fungus in the genus Schizophyllum. As the general information about this type of mushroom, Schizophyllum commune mushroom is known as the white-rot fungi. Based on this research, the lignocellulose content in rice husk which are hemicellulose, lignin, and cellulose were determined by extraction of biomass by using benzene, extraction of hemicellulose by sodium hydroxide, and extraction of lignin by sulphuric acid. By using specific mathematical formulae, hemicellulose, lignin, and cellulose were obtained as 7.73%, 81.99%, and 5.55% respectively. Next, effects of different amount of medium which were water, rice bran, and calcium carbonate added on mycelium growth rate were analysed by One Factor at Time, OFAT. From the varied amount, the rapid mycelium growth was produced from the amount of water added at 70 ml, rice bran added at 50 g, and calcium carbonate at 2 g. Regarding to third objective, the mycelium yield obtained from different content of substrate composition was evaluated by weighing substrate composition that produce the rapid mycelium growth rate. Based on the result obtained, substrate composition that contained 2 g of calcium carbonate was the highest mycelium yield which is 10.88 g then followed by yield from substrate composition that contained 50 g of rice bran added which is 10.67 g, and substrate composition that contained 70 ml of water added which is 10.43 g. Lastly, optimum of substrate composition formulation was contained 70 ml of water content, 50 g of rice bran, and 2 g of calcium carbonate added.en_US
