Abstract: Acne vulgaris is a chronic inflammatory diseases caused by normal skin flora. A patient with this diseases are commonly being prescribed with antibiotics. However, due to emergence of multiple bacteria resistance toward antibiotics has raised a great concern in pharmaceutical industry. In the same time, a herbs known as Clinacanthus nutans leaves (C. nutans) or Belalai gajah leaves have been used for centuries in treatment of skin diseases. Therefore, the purpose of this project is to investigate the potential of optimized extract yield of this herbs for antibacterial activity towards acne vulgaris causing bacteria. The dominant microorganism isolated from acne vulgaris patient was found to have characteristic of a rod shape, non-spore and gram positive bacilli bacteria. The optimization of methanolic extract with temperature, extraction time, and solvent to water ratio (s/w)% to produce optimum result where the response was maximized by utilizing the temperature of 60 degree C, time of 20 minutes, and 80% (s/w) ratio yield TPC of (27.58±0.0005mg GAE/g). The antibacterial susceptibility test was done using well diffusion method for optimized methanolic extract of C. nutans and resulted inhibition zone of (9±0.03mm). Hence, these result can be one of supported scientific data that shows the potential of methanolic extract as antibacterial agent for treatment of acne vulgaris.
