Title: Statistical optimisation for the effect of Calcium Phosphate precipitation on the growth of MCF-7 cancer cell

Abstract: Breast cancer is one of the many cancer cases that caused death to women. MCF-7 breast cancer cells have been widely used in the research of cancer. The aim of this study is to investigate the growth condition of MCF-7 cancer cell using calcium phosphate precipitation method. The growth curve profile of MCF-7 cancer cells was constructed to determine the exponential phase. The cells at exponential phase was used to screen the significant parameters; concentration of phosphate, concentration of calcium and cell exposure time that affected the growth condition using Two Level Full Factorial Design. The screened parameters were subjected for optimization study using Box-Behnken Design to obtain the optimal value for growth condition. After a lag period of about 76 hours, these cells grew exponentially until 194 hours with specific growth rate at 0.0258 and doubling time of 27 hours. The best growth condition for MCF-7 cells is at 5 M concentration of calcium, 2 M concentration of phosphate, and 6 hours of cell exposure time with 39 ± 2.309% of viable cells. The results obtained from this study can be used for future research on MCF-7 cancer cells transfection using calcium phosphate co-precipitated with DNA and can be further used in the study for breast cancer immunotherapy.
