dc.description.abstract | Anthracnose disease is a fungi or bacteria disease that is caused by several microorganisms. It is a devastating disease that established on panicles of the mango and then virtually attacks them. Mangoes are usually influenced by this infection when they are young. The side effects show up particularly amid and in the wake of aging of the tainted, develop (mature) fruit and the symptoms are clear enough to be seen with naked eyes. The aim of this study to isolate and identify the microorganism (fungi or bacteria) from the mango with anthracnose disease. The mango with anthracnose disease symptom are used as a sample. The black spot on the sample are cut into a small piece and transfer onto the Potato Dextrose Agar (PDA). Several colony of microorganism are on PDA plate after a week of incubation at room temperature. Each colony of sample undergo streak plate to obtain a pure culture. The selected microorganism were classified as Isolate 1, Isolate 2, Isolate 3, Isolate 4, Isolate 5 and Isolate 6. After that, Colony-PCR are proceed to get the DNA from the cell from each Isolate. DNA from Colony-PCR are used as a template for the amplification of Polymerase Chain Reaction (PCR) for molecular identification by using 16s rRNA with primer 27F and 1492R as a target sequence. The PCR product were analysed by Gel Electrophoresis and visualized under UV light of Gel Documentation. Prepared PCR product are sent to the DNA sequencing and In Silico Analysis provider. Phylogenetic tree are constructed and the microorganism are identified which are Citrobacter sp., Pseudomonas sp. and Pantoae sp. | en_US |
