dc.description.abstract | This research is discuss on the anodized al oxide for DNA detection by using Taguchi method. One step anodizing is carry out in the mixture electrolyte where phosphoric and oxalic acid are used. Fixed parameter in this studies is voltage, concentration used and also time while noise factor is volume of the electrolyte and different power supply is use. The anodizing process will be cary out in 30 , 45 and 60 minutes. After getting the coating surface of the anodizing part , polarization test were done to studied the corrosion behavior which is polarization resistance that will be optimize later to get best three sample by Taguchi method. For sample 1 with 0.5M , 30 min and 20V , second is 0.4M, 40min and 30V and lastly third sample with 0.3M ,60min and 40V. These three sample is then undergo DNA immobalization with 10μM DNA with amine-terminated group and leave it in 4h at RT . After that ,Cyclic voltammetry is used to detect DNA by measure the immobilization current (Iimmo) . The voltage used is between -1.5 to 1.5 V with the rate of 0.100 V/s .Potassium Hexacyanoferrate III[ K3Fe(CN)6] in 0.1 KCl used as electrolyte bath.From the result , the anodized part with DNA immobalized have higher immobalized current than polarized part with DNA immobalized . Scanning Electron Microscope (SEM) is to observe the surface morphology of the anodized and polarize part . The result show anodized part have more particle and dispersed around than the polarize part. | en_US |
