Abstract: | The optimization of cellulase enzyme production was studied in solid state fermentation (SSF) on sugarcane bagasse by Aspergillus niger. Three parameters which are moisture content, fermentation period and inoculums size that influenced the production of cellulase were studied using 2-level full factorial design for screening process and the significant parameters obtained are fermentation period and inoculums size were further analyzed using Response Surface Methodology (RSM). Central Composite Design (CCD) was used to develop a statistical model for the optimization of process parameter used. Data that obtained from RSM on cellulase production were subjected to the Analysis of Variance (ANOVA) and analyzed using a model equation resulted in the optimized process parameters. The optimize conditions were obtained are at day 5 of fermentation period and inoculum size of 4x107 spores/g substrate. The maximum cellulase production in term of enzyme activity was 0.3093 IU/mL. Therefore, the statistical analysis performed was successfully developed a model that was reliable for optimization of cellulase enzyme production. |
